紫茎泽兰热激蛋白70基因的克隆与序列分析

摘要:

热激蛋白70(HSP70)与植物的抗逆性密切相关。根据其他植物hsp70的保守性区域设计特异性引物,采用RTPCR方法从紫茎泽兰Ageratina adenophora中克隆到hsp70基因全长cDNA序列。序列分析结果表明,该cDNA包含1个 1 938 bp的开放阅读框(GenBank登录号:FJ598132),共编码645个氨基酸,相对分子质量为70.58 kDa,命名为hsp70.58。该蛋白包含真核生物HSP70高度保守的三个家族标签和末尾基序EEVD。运用实时定量PCR 分析,探测到hsp70.58在mRNA水平上皆能被高温和低温诱导表达,说明该基因属于诱导型hsp70基因。

Abstract:

Heat shock protein 70 (HSP70) plays an essential role in plant stress resistance. According to the conserved regions of hsp70 from other plant species, a pair of specific primers were designed. A fulllength cDNA of hsp70 gene from Ageratina adenophora was cloned by the technique of RTPCR. Its ORF (open reading frame) (GenBank locus: FJ598132) named hsp70.58 was 1 938 bp. It encoded a polypeptide of 645 amino acids with an estimated molecular mass of 70.58 kDa and contained the three highly conserved HSP70 family signatures and Cterminal pentapeptide EEVD. Relative realtime RTPCR was used to analyze the expression of hsp70.58 gene after heat shock and cold shock, respectively. The results showed that mRNA expression of hsp70.58 was significantly upregulated after temperature stress. It indicated that this gene belonged to hsp70 inducible type.
