棉花GhSRK2D基因的分子克隆及其功能分析

Molecular Cloning and its Functional Analysis of GhSRK2D Gene from Cotton

摘要:

SnRK2类蛋白激酶在植物抵御非生物胁迫上起到非常重要的作用,可将其作为候选基因,通过转基因方式对作物进行耐旱性改良。 本研究利用RACEPCR技术从陆地棉分离到一个SnRK2同源基因GhSRK2D。生物信息学分析表明GhSRK2D属SnRK2sⅢ亚家族;GhSRK2D蛋白含有N豆蔻酰化位点,跨膜结构域和5个核心的丝氨酸残基位点;另外, GhSRK2D蛋白的C末端保守结构也包含Osmotic Box、SnRK2 Box 和 ABA Box。 Southern杂交分析GhSRK2D基因在Y18生态型棉花基因组中含有两个拷贝;Realtime PCR分析显示GhSRK2D在棉花不同组织器官中均有表达,在叶器官中表达量最高;且受ABA、高盐、高渗(甘露醇)及低温胁迫的诱导。这些结果预示了棉花GhSRK2D基因可能在非生物胁迫逆境下参与抗逆相关生理过程发挥重要作用。

Abstract:

The protein kinases SnRK2s play important roles in abiotic stress response in plants and likely act as candidate genes for improving stress tolerance in crops through transgenic approaches. In this study, SnRK2 homologous, GhSRK2D was isolated from the Y18 upland cotton via RACEPCR. Bioinformatics analysis indicated that GhSRK2D belonged to SnRK2sⅢ subfamily, containing the Nmyristoylation sites and transmembrane domains, and 5 core serineresidues sites. Additionally, Osmotic box, SnRK2 box and ABA box were also found in the end of Cterminal conserved domain of protein GhSRK2D. Southern blot results showed that GhSRK2D gene had 2 copies in cotton Y18 ecotype genome. Realtime PCR showed that GhSRK2D was expressed in various cotton tissues and organs, the highestlevel expression of GhSRK2D gene was identified in leaf organs. GhSRK2D was also  induced by ABA, high salt, hypertonic (mannitol) and low temperature treatments.  These results suggested that GhSRK2D was likely involved in abiotic stress physiological processes in cotton.
