Abstract:
Based on the growth of Saccharopolyspora spinosa in both solid and liquid medium, a high throughput method was established to screen high spinosadproducing strains with 96well plates. The results showed that applying the sandwich covers for 96well plates could meet the requirement of bacteria filtration and oxygen supply for the growth of S.spinosa. S.spinosa grew well in both 96well plate and slant culture. The optimum seed growing time was 60 h; the broth content of each well was 0.4 mL. Comparing with the traditional shake flask screening platform, this method could significantly improve the screening efficiency of high spinosadproducing strains.

摘要:
考察了刺糖多孢菌在96孔板固体培养基和液体培养基中的生长情况,建立了96孔板高通量筛选多杀菌素高产菌株的方法。结果表明:96孔板三明治盖能同时满足过滤杂菌和刺糖多孢菌生长耗氧所需,刺糖多孢菌在96孔板固体培养基中的生长情况与传统斜面培养相当,种子培养最适种龄为60 h,每孔发酵培养基装液量为0.4 mL。与传统摇瓶发酵筛选平台相比,该方法能显著提高多杀菌素高产菌株的筛选效率。
