摘要:

为了提高毕赤酵母(Pichia pastoris)中重组植酸酶的表达,在来源于Pichia stipitis的低氧诱导启动子PsADH2控制下,进行了透明颤菌血红蛋

白基因vgb在重组P. pastoris中与植酸酶的共表达研究。结果表明,PsADH2启动子在低氧条件下被诱导启动,表达的血红蛋白VHb对P. pastoris

宿主细胞生长无明显影响,但使重组P. pastoris中共表达的植酸酶APPA活性提高了3倍。

Abstract:

To improve the production of a recombinant phytase APPA in Pichia pastoris under O2 limitation, the PsADH2 promoter from

Pichia stipitis was employed to control the expression of Vitreoscilla hemoglobin (VHb) encoding gene (vgb) in P. pastoris.

This promoter was activated under O2 limitation in P. pastoris. The expressed VHb enhanced the phytase APPA activity to 3

fold higher than that of VHb-free cell under O2 limitation, but no obvious effect on cell growth was observed.
