Study of Immature Embryo in vitro Culture in Reciprocal Cross Hybrid Between Commom Wheat and Cultvate Barley

小麦与大麦正反交杂种幼胚离体培养效果研究

Abstract:

In order to explore the effect of immature embryo in vitro culture in reciprocal cross hybrid between commom wheat and cultivate barley, two ways of immature embryo rescue,  embryo culture and embryo-callus culture, were compared on their effects. The result showed that there were 28.71% embryos germinated, 13.04% embryos rescue succeeded, 28% transplant survived with embryo culture when common wheat Shi4185×cultvate barley W30 immature embryo in vitro cultured. There were 38.83% embryos germinated, 17.51% embryos rescue succeeded, 34.21% transplant survived with embryo culture when cultivate barley W30×common wheat Shi4185 immature embryo in vitro cultured. There were 16.67% frequency of callus induction,  88.41% transplant survived with embryo-callus culture when commom wheat Shi4185×cultvate barley W30 immature embryo in vitro cultured. There were 10.45% frequency of callus induction, 89.29% transplant survived with embryo-callus culture when cultvate barley W30×commom wheat Shi4185 immature embryo in vitro cultured. It was concluded that a large number of healthy regeneration plant was gained with embryo-callus culture. This is a ideal way of immature embryo in vitro culture in reciprocal cross hybrid between commom wheat and cultvate barley.

摘要:

为了探讨普通小麦与栽培大麦正反交杂种幼胚离体培养效果,采用幼胚直接成苗和幼胚-愈伤组织成苗两种培养方法进行杂种幼胚离体培养。结果表明,采用幼胚直接成苗培养法,小麦石4185×大麦W30胚培出苗率为28.71%,胚拯救成功率为13.04%,移栽存活率为28%;大麦W30×小麦石4185胚培出苗率为38.83%,胚拯救成功率为17.51%,移栽存活率为34.21%。采用幼胚-愈伤组织成苗培养方法,小麦石4185×大麦W30胚培出愈率为16.67%,移栽存活率为88.41%;大麦W30×小麦石4185胚培出愈率为1045%,移栽存活率为89.29%。说明通过幼胚-愈伤组织成苗培养法可以获得大量健壮再生植物,是普通小麦与栽培大麦正反交杂种幼胚离体培养的理想方法。
