96孔板高通量筛选多杀菌素高产菌株的研究

Studies on High Throughput Screening for High Spinosad-producing Strains with 96-well Plates

摘要:

考察了刺糖多孢菌在96孔板固体培养基和液体培养基中的生长情况,建立了96孔板高通量筛选多杀菌素高产菌株的方法。结果表明:96孔板三明治盖能同时满足过滤杂菌和刺糖多孢菌生长耗氧所需,刺糖多孢菌在96孔板固体培养基中的生长情况与传统斜面培养相当,种子培养最适种龄为60 h,每孔发酵培养基装液量为0.4 mL。与传统摇瓶发酵筛选平台相比,该方法能显著提高多杀菌素高产菌株的筛选效率。

Abstract:

Based on the growth of Saccharopolyspora spinosa in both solid and liquid medium, a high throughput method was established to screen high spinosadproducing strains with 96well plates. The results showed that applying the sandwich covers for 96well plates could meet the requirement of bacteria filtration and oxygen supply for the growth of S.spinosa. S.spinosa grew well in both 96well plate and slant culture. The optimum seed growing time was 60 h; the broth content of each well was 0.4 mL. Comparing with the traditional shake flask screening platform, this method could significantly improve the screening efficiency of high spinosadproducing strains.
