烟草突变体库的创建策略及其应用

Abstract:

Tobacco (Nicotiana tabacum) is one of the most important cash crops in the world and it is also a model plant for biotechnology research. At present, tobacco mutants has been widely used in functional genomics study and breeding, which will be further promoted by constructing an efficient and large-scale tobacco mutant library. This paper reviews the applications of chemical mutagenesis, ion radiation-induced mutagenesis and insertion mutagenesis in mutant library construction. In addition, considering the character of tobacco genome, new strategies of  tobacco mutant library construction were described in combination with new methods for functional genomics study, including whole genome overexpression strategy based on Gateway technology and EST-RNA interference strategy based on RMHR technology. How to utilize them to study tobacco functional genome is also discussed in this paper.

摘要:

烟草(Nicotiana tabacum)是重要的经济作物及研究植物生命活动的模式植物。目前烟草突变体已广泛应用于烟草功能基因组研究及遗传育种,大规模高效烟草突变体库的创建将进一步推动烟草功能基因组研究,加快新品种的培育。综述了化学诱变、电离辐射和插入标签法在烟草突变体库创建中的应用;此外,针对烟草基因组的特点,结合近年来一些植物功能基因组研究的新技术、新方法,提出了烟草突变体库创建的新策略,即基于Gateway技术的烟草全基因组过量表达策略和基于RMHR技术的烟草EST-RNA干涉策略,并就如何应用这些突变体库进行烟草功能基因组研究进行了探讨。
